well i just slim'd my xbox so i cant try it, but if i can remember the xbox drive did NOT have a lip to hold the game in the tray so it "might" work i wouldnt recommend it.
I was under the impression as well that the ps2 would eventually have troubles with the laser if it was on its side. I would assume that the xbox would be the same way, including the 360.
The xbox DVD drive does have a lip and I started a game on its side. I doubt it was made to be played like that though. If you want to play on its side, you should consider upgrading the HDD and play from there.
the xbox gets hot enough as it is, blocking a air vent might not be the best thing to do, but if you watch your temps and maybe do the 80mm fan upgrade you will probably be fine.
i wouldnt recommend at all, its not like the ps2 where it can be use on the side. if the xbox was used on the side, i would think it would over heat and get hot. also you would be doing harm to the dvd drive on it. making it work harder.
You should have no problem sitting it on it's side. FYI, Your console will last twice as long then it would on it's flat side. If you go that route, it MAY generate alot of heat. Just try it and see for your self.
